strange, i can't find a combination of enemys that actually make things hard in ep4.
I mainly played ep1 but i have seen ep2 and ep4 so i will say ep2 is the hardest as it has lot's of enemys with 1 hit KO moves.

deffo ep2 but for all those annoying Sinows and Mr Gibbles
But saying that i havent really played ep4.
