You answered your problem in itself. The first time you fight Ampulus Umbra, you have to have beaten BOTH Castor and Pollux. Whichever you defeat last, you enter the Ampulus Umbra battle. So in order to fight him, you'll have to complete the hunter storyside and fight it with a hunter.
