I don't think so - but I tend to kill all of the Forest enemies because it's easy experience. And in Ultimate, I'll take as much of it as I can get.
You could always try it and see.
I know that you don't have to kill all of the Caves enemies, so logically, you'll not have to kill all of the Forest enemies.
